Effect of Red Ginseng Extract (RGE) on Learning and Memory Impairment Animal Model SAMP8 - III. Feeding Effect of RGE on Neurotransmitters and Their Metabolites in SAMP8 Brain

Abstract
This study was designed to investigate the effects of red ginseng extract (RGE) extracted with 80% ethanol on oxygen radicals and their scavenger enzymes of learning and memory impairments SAMP8 (female). Ginseng (Panax ginseng C.A. Meyer) is one of the most plant drugs which has been used as a medicine in Asia. Senescence-accelerated mouse (SAM) P8 were fed basic diets (control group) and experimental diets (RGE group) with 250 §·/§¸ BW of RGE for 8 months. Passive avoidance tests such as acquisition trial and retention test in RGE group were significantly higher (10% and 15%) than those in control group. Acetylcholinesterase(AChE) activity was significantly increased about 45% by long-term administration of RGE compared with control group. Neurotransmitters such as dopamine (DA) and serotonin (5-HT) and their metabolites such as 5-hydroxyindole acetic acid (5-HIAA) in RGE group were significantly lower (15¡­20%) than those in control group, but their metabolities such as homovanillic acid (HVA) showed almost no change in RGE group. HVA/DA ratio of RGE group was higher (25%) than that of control group, but 5-HIAA/5-HT ratio of RGE group was lower(10%) than that of control group. Monoamine oxidase B(MAO-B) acitivity in RGE group was significantly inhibited about 10% compared with control group. These results suggest that long-term administration of RGE may be activated retention test, acetylcholinesterase activity and ratio of metabolites on neurotransmitters, resulting in a greater prevention of learning and memory impairments such as Alzheimer-type dementia.
